What is Epithalon?

Epithalon is a synthetic tetrapeptide, Ala-Glu-Asp-Gly, whose sequence was derived from the amino acid composition of Epithalamin, a bovine pineal gland extract. Distinguishing the two is essential when reading this literature: Epithalamin is a complex peptide extract, whereas Epithalon is a defined synthetic tetrapeptide, and the two have largely separate evidence bases. The most-cited proposed mechanism is telomere-related — addition of Epithalon to telomerase-negative human fetal fibroblast cultures was reported to induce expression of the telomerase catalytic subunit, enzymatic telomerase activity, and telomere elongation, with a follow-up reporting that treated fibroblasts underwent approximately ten additional passages beyond the control replicative limit.

A 2025 independent study reported dose-dependent telomere elongation in normal epithelial and fibroblast cells via hTERT and telomerase upregulation, and telomere extension in breast cancer cell lines via the Alternative Lengthening of Telomeres pathway, the ALT contribution being largely cancer-cell-specific; note that this paper carries a published correction. A second proposed mechanism involves direct interaction with nucleic acids, with reports that short regulatory peptides bind the DNA double helix and modulate gene transcription, though structural and biophysical characterization of this interaction remains limited. In rodent models, Epithalon has been reported to reduce chromosome aberration frequency in bone marrow cells and to downregulate HER-2/neu transcript expression in mammary tumors of transgenic mice.

Importantly, the frequently asserted claim that Epithalon restores melatonin secretion is not supported by direct measurement: an independent perifusion study found that Ala-Glu-Asp-Gly had no significant effect on melatonin secretion by pineal glands from either young or old rats, and did not modify stimulated melatonin release. The overall mechanistic picture remains poorly resolved.

What is the difference between Epithalon and Epithalamin?+

This distinction is essential when reading the literature. Epithalamin is a complex extract of bovine pineal tissue; Epithalon is a single defined synthetic tetrapeptide (Ala-Glu-Asp-Gly) derived from its amino acid composition. Most of the human observational literature concerns Epithalamin, while the telomerase and cell-culture work concerns Epithalon. Findings from one should not be attributed to the other.

What is the proposed mechanism?+

The most-studied hypothesis is induction of hTERT expression and telomerase activity with consequent telomere elongation, demonstrated in human cell culture. A secondary hypothesis involves direct peptide-DNA interaction modulating gene transcription, though this is not well characterized structurally. Recent review work notes that physicochemical and structural investigation of the peptide remains quite limited.

Does Epithalon affect melatonin?+

This is frequently claimed but is not supported by direct measurement. An independent perifusion study found that Ala-Glu-Asp-Gly produced no significant effect on melatonin secretion by pineal glands from young or old rats across the concentrations tested, and did not alter stimulated release. Melatonin-related claims should be treated as unsupported by the available direct evidence.

Are there human clinical trials?+

No. A ClinicalTrials.gov search returns no registered studies of the synthetic tetrapeptide. Human studies commonly cited in this context used Epithalamin, the bovine pineal extract, and were non-blinded, non-placebo-controlled, unregistered, and conducted by the institutions that developed the preparations.

How should the geroprotection literature be interpreted?+

With considerable caution. The rodent lifespan data are more limited than commonly represented — in the principal SHR mouse study, mean life span was not affected, with effects confined to maximum life span and the longest-lived decile. Most in vivo work originates from the institutions that developed the compound, replication outside those groups is sparse, and no registered human trials exist. Claims of anti-aging or life extension in humans are not supported by the published evidence.

What are the handling and stability considerations?+

As a short, highly polar lyophilized tetrapeptide it is hygroscopic; equilibrate to room temperature before opening. Reconstitute in sterile or bacteriostatic water with gentle swirling rather than vortexing, and aliquot to avoid repeated freeze-thaw cycles. Confirm identity and purity by HPLC and mass spectrometry on receipt.

Epithalon is not approved by the U.S. Food and Drug Administration for any indication and is not authorized for human use in the United States. FDA has placed Epitalon in Category 2 of the bulk drug substances nominated under sections 503A/503B of the FD&C Act — substances that may present significant safety risks — stating that compounded drugs containing epitalon may pose immunogenicity risk for certain routes of administration due to potential aggregation and peptide-related impurities. FDA's Pharmacy Compounding Advisory Committee reviewed Epitalon-related bulk drug substances in July 2026 for possible inclusion on the 503A Bulks List; advisory committee recommendations are non-binding and this status is live and may change, so verify against FDA's current published position rather than any secondary source.
